In the season 6, we've learned how the Children of the Forest made the White Walkers using dragonglass.
This scene :
Then, we can conclude that the effect of dragonglass on White Walkers is clearly tied to how the Children of the Forest made the Night King.
But what's more interesting is why they've created him : they were at war with the humankind.
Leaf : "we've been slaughtered, our sacred trees cut down..."
Sounds familiar ?
it's a classic trope in modern fantasy : the evils of the modern world: ugliness, depersonalization, and the separation of man from nature.
Avatar, Lord of the Rings, Dances with wolves (my favorite), and many more have used this trope.

I strongly believe that the saga is about the balance between Ice and Fire (in a symbolic way, not by magic...)
The fate of Ice (white walkers), he's tied to the fate of Fire (dragons / targaryens). One can not survive without the other. One will ever oppose the other.
The Fire can represent in an allegoric way how the human can be monsters : the mad king, tagaryens dynasty for all their work, war of the seven kingdoms, the first men vs the childrens of the forest, etc.
In this time, the NK (Ice) was a necessary evil. Leaf confessed it.
And things eventually went south went the NK turned rogue, out of control...
But now, as long as long as a targaryen remains, there will be "Fire". This family personifies how madness turns men into monsters.
And the Ice (death of men) will oppose the Fire (madness of men) as it was the original purpose.
Please, note how they mirror each other : both NK and targaryens were men before becoming monsters
The shard of dragonglass used to create the NK can personify this link.
(Dragonglass come for from the fire inside earth. Regarding the dragons, the valyrians claimed that dragons sprang forth from the Fourteen Flames, a ring of volcanoes on the Valyrian peninsula.)
They're just tied together.
